Annexin V 是检测早期细胞凋亡的最灵敏指标之一。正常细胞中,磷脂酰丝氨酸 (PS)只分布在细胞膜磷脂双分子层的内侧,而在细胞凋亡早期,细胞质膜内侧的磷脂酰丝氨酸发生外翻,暴露于细胞质膜的外侧。Annexin V是一种与磷脂酰丝氨酸有高度亲和力的Ca2+依赖性磷脂结合蛋白,因此能够利用Annexin V与细胞外侧暴露的磷脂酰丝氨酸相结合的性质,检测细胞的早期凋亡。将 Annexin V进行绿色荧光 (FITC) 标记,以标记了的Annexin V-FITC 作为探针,利用流式细胞仪或荧光显微镜可检测细胞凋亡的发生。碘化丙啶 (PI)是一种核酸染料,它不能透过具备完整细胞膜的正常细胞和早期凋亡细胞,但能够透过凋亡中晚期的细胞和坏死细胞的细胞膜而将细胞核染色,在特定激发光条件下呈现红色。
将 Annexin V-FITC与 PI 匹配使用,就可以将处于不同凋亡时期的细胞区分开来。在双变量流式细胞仪的散点图上,若横坐标表示Annexin V信号,纵坐标表示PI信号,则左下象限 (Annexin V-/PI-) 代表活细胞;右下象限(Annexin V+ /PI-) 代表早期凋亡细胞;右上象限(Annexin V+ /PI+)代表凋亡晚期细胞和坏死细胞;左上象限(Annexin V-/PI+) 被认为是许可范围内的检测误差。
